水素原子をMe基で置換することにより，conformationの変化やDIを惹起して劇的な活性の向上が見られることがあります．このようなMe基はMagic Methyl (group)と呼ばれ，LEやLLEなどの重原子あたりのリガンド効率に大きく寄与します。
